In this section, you can find condensate-relevant methods developed and/or utilized by the labs in Dresden.

  • The EMBO course on Methods for Studying Phase Separation in Biology was held in March 2019 in the MPI-CBG, Dresden. A manual that contains the methods for in vivo and in vitro investigation of the liquid-liquid phase separation phenomenon was developed. Below are the methods published in that manual:
    • Determining the intracellular pH of HeLa cells
    • Determining the in vivo saturation concentration using quantitative fluorescence microscopy
    • Determining the in vitro saturation concentration using fluorescence microscopy
    • Analysis of client partitioning into polymeric coacervates
    • Characterization of protein-based phase separation parameters using temperature-controlled microscopy
    • Measuring material properties of phase separated protein droplets with dual-trap optical tweezers
    • Quantification of molecular dynamics in phase separated protein solutions using fluorescence correlation spectroscopy
    • Fluorescence recovery after photobleaching (FRAP) in protein condensates
  • Standard Operating Procedure (SOP) protocols have been developed in Dresden to standardize methods used in the field. Currently available SOPs are listed below:
    • FUS protein purification
    • PGL3 protein purification
    • XMAP215 protein purification
